What Is Liquid Filled Hard Capsules?

Liquid-filled hard capsules are a type of pharmaceutical dosage form that combines the benefits of traditional hard capsules with the ability to contain liquid formulations. These capsules consist of two main components: a hard outer shell, typically made from gelatin or a suitable vegetarian alternative, and a liquid fill that can include a variety of substances such as oils, suspensions, or solutions of active pharmaceutical ingredients (APIs).

The primary advantages of liquid-filled hard capsules include improved bioavailability of the active ingredients, as the liquid form can enhance absorption in the gastrointestinal tract. They also allow for the encapsulation of a wider range of formulations, including those that are sensitive to moisture or require specific delivery mechanisms. Additionally, liquid-filled capsules can mask the taste of unpleasant medications, making them more palatable for patients.

Types of Liquid Filled Hard Capsules
Liquid Filled Hard Capsules
Liquid Filled Hard Gelatin Capsules

Liquid Filled Hard Capsules

Hard gelatin capsules
The gelatin used in the manufacture of most common capsules is obtained from collagenous material by hydrolysis. Gelatin is a natural, safe, non-allergenic, clean, and economical ingredient. The two-piece hard gelatin capsule is available in a range of sizes; from largest to smallest, these sizes are 000, 00el, 00, 0xel, 0el+, 0el, 0, 1el, 1, 2, 3, 4, 5.

HPMC capsules
HPMC capsules are stable at low humidity levels, have low moisture content (3–8%), and low static charge. These natural capsules are available from size 00 to 4. HPMC capsules are suitable for highly reactive molecules (because they have no cross-linking reactions). Compared to hard gelatin, HPMC is more suitable for moisture-sensitive products, hygroscopic products, and for low relative-humidity applications. HPMC is not of animal origin and does not pose a risk of contamination with organisms that cause bovine spongiform encephalopathy (BSE) or transmissible spongiform encephalopathy (TSE). HPMC capsules are used in a wide range of OSD pharmaceuticals as well as nutraceuticals, dietary supplements, and herbal products, due to the vegetarian nature of HPMC.

Fish gelatin capsules
Marinecaps are made from fish gelatin and do not pose risk of BSE or TSE. They are preferred for filling marine supplements such as EPA [eicosapentaenoic acid]-rich fish oil.

Starch capsules
Starch capsules are made from potato starch. Their dissolution is pH independent, and they are suitable for enteric coating. The moisture content of starch capsules ranges between 12–14% w/w, with more than 30% being tightly bound (1).

Pullulan capsules
These vegetarian capsules are made from tapioca, which is naturally fermented into pullulan. They provide a high barrier to oxygen.

Polyvinl acetate (PVA) capsules
Capsules made from PVA can be used for filling insoluble drugs dissolved in polyethylene glycol (PEG) 400. PEG 400 when used as single vehicle is not compatible with other hard capsules. The oxygen permeability of PVA is low, resulting in a high barrier to oxygen.

Liquid-filled hard capsules (LFHC)
Two-piece hard capsules made of either gelatin or HPMC can be used for filling and band sealing non-aqueous liquid, paste, suspension, hot melts, and other vehicles that melt up to 70 °C and flow easily. LFHC can also be filled with tablets, pellets, or other capsules as combination fill.

Soft gelatin capsules (SGC)
SGC have soft, globular, gelatin shells somewhat thicker than that of hard gelatin capsules. The gelation is plasticized by the addition of glycerin, sorbitol, or a similar polyol. It may contain preservative to prevent the fungal growth. Large-scale production methods are generally required for the preparation of SGC.

How To Fill Gelatin Capsules With Liquid?

Preparation of empty capsules and liquid fills
The operator loads empty capsules and liquid fills into their hoppers. Capcn-l30 is capable of handling capsule sizes ranging from #00 to #5. Aside from liquids, the equipment can also fill many other materials, such as semi-solids, suspensions, pastes, and waxy substances.

Capsule feeding and separation
During this stage, the empty capsules are rectified and transferred to the segments in the vertical position. When each capsule is fed into the segment pore, they are ready for the next step - separation.

The vacuum system plays a key role in capsule separation. When the capsules are separated, the bodies fall into the bores of the lower segment, while the caps remain in the bores of the upper segment.

Liquid dosing and filling
After separation, the lower segment holding empty capsule bodies moves to the filling station. Liquid fills are pumped into the filling station by a peristaltic pump. The pump enables precise liquid dosing without leaking, dripping or air bubbles.

Capsule closing
Now the lower segment carrying filled capsule bodies rotates to the closing station. As with conventional capsule filling machines, the upper segment is lined up with the lower one. A row of pins pushes the bodies up from the bottom of the lower segment to close the capsules.

Filled capsule transfer
In the above four steps, any capsule that fails to separate or close is rejected by vacuum. In this stage, the closed capsules are pushed out of the segments into the holes of a rotary disc. All segments are then cleaned by vacuum and go back to the first place for the next circle.

Filled capsule sealing
The sealing mechanism is mounted under the slat conveyor. It comprises two sealing rollers, two gelatin bowls, a water tank, and a gelatin tank. By the way, two gelatin bowls and the gelatin tank are equipped with heaters to maintain the temperature of the gelatin.

Sealed capsule drying
The sealed capsules are then moved to the drying carriers from the slat conveyor and transferred into the drying box. The drying box uses filtered indoor air at a constant room temperature to dry the capsules on the carriers.

Application of Liquid Filled Hard Capsules

Combination therapy
It is helpful for illnesses needing different drugs. Because it allows the combination of several active ingredients in a single dose.

Oral drug delivery
Liquid-filled capsules are used in a wide range of pharmaceutical drugs. It could be one of those with particular controlled-release specifications and low solubility. These capsules have liquid or semi-solid formulations inside them. That's why liquid capsules allow for better bioavailability and high control over drug release kinetics.

Pediatric and geriatric medications
Liquid-filled capsules can be an excellent choice, especially for children and older patients who have trouble swallowing solid pills or capsules. Liquid-filled capsules can be a great alternative.

Functional ingredients
These capsules are a favorite choice to deliver functional ingredients like probiotics, vital oils, or food additives in another way.

Flavor enhancers
In the food industry, liquid-filled capsules are commonly used for flavoring and aromas for food products, including beverages, spices, and sweets.

Agriculture: Pesticides and fertilizers
Agriculture commonly uses liquid-filled capsules to protect fertilization. Most of the time, pesticides are controlled.

Vitamins and minerals
Liquid-filled capsules are commonly used to encapsulate vitamins, minerals, and other nutritional supplements. Improved bioavailability and absorption may result from it.

Omega-3 fatty acids
Due to their oxidation sensitivity, omega-3 supplements, often made from fish oil, are usually supplied in liquid-filled capsule form.

Herbal extracts
Liquid-based capsules help in delivering plant-based supplements, botanicals, and herbal extracts.

Process of Liquid Filled Hard Capsules

Raw material melting & coloration
The raw material that meets the acceptance test requirements is brought into the manufacturing area.

Raw material contained in the hopper is automatically weighed in the container.

Now, the weighted material will be shifted from the container to the melting tank.

Purified water is added to the melting tank before finally starting the melting process.

After initiating the melting, the mixture gets stirred to create a uniform jelly.

In this step, the colorant can be added to the colored capsules.

Capsule molding & cutting
In the capsule production room, the capsule manufacturing machines receive the colored jelly.

The pins are then dipped into the jelly mixture. Molding pins are then shifted to the drying unit.

The drying process will be at a constant temperature and humidity. After the drying process, these are sent to the next section.

Now, the dried caps and bodies are removed from the molding pins.

The caps and bodies will be trimmed to an appropriate length and then pre-locked.

Next, an immediate visual inspection of all the capsules will be done. The light is flashed on the rotating capsules, and the digital images are captured with cameras. Besides, image processing technology is used to detect abnormal shapes.

In the last step, the conformed capsules will be packed into the containers.

Printing, inspection & packaging
Now, the printing of the capsules started for the product identification purpose.

The specified design is printed in the capsules by utilizing the ink and printing machine of the company.

An alternate option can also be used for printing without ink. In this part, you can use the laser printing machine.

Packaged capsules are stored in the product warehouse with controlled temperature and humidity.

Shipping tests & shipment
The companies can do some tests before the shipment of the right capsule products. Capsules are generally tested for dimensions, appearance, moisture content, separation, and more.

FAQ

Q: What are liquid-filled capsules?

A: A hard gelatin capsule filled with liquid medication is made with two halves - the rim of one half is smaller in diameter than the other, so the other half's larger rim fits over it and seals the capsule. The two capsule halves are manufactured separately, then later filled using different machinery.

Q: What are the benefits of liquid filling capsules?

A: Liquid-filled capsules are extremely quick compared to powder-filled capsules or tablets." The bioavailability of poorly water-soluble drugs has been proven to be enhanced by the use of liquid-fill technology1,2, while it continues to present an obstacle for tablet formulations.

Q: How do you take liquid-filled capsules?

A: Instead of crushing, chewing, or dissolving your softgels, swallow them with their coating intact, unless your doctor or pharmacist tells you to take them differently. Softgels contain liquid, and their outer coating is designed to dissolve in your stomach or small intestine.

Q: Do liquid-filled capsules work faster?

A: In general, your body will benefit more from an appropriate dose of either type of liquid-filled capsule than it will from tablets. That's because liquid ingredients are more easily and quickly absorbed into the body. In other words, liquid gels work faster than tablets.

Q: How do you swallow a liquid capsule?

A: Bend the head forward by tilting your chin slightly toward your chest. Swallow the capsule and the water with the head bent forward.

Q: What are hard capsules for liquid?

A: Liquid-filled hard capsules (LFHCs) are typically composed of a shell of gelatin or hydroxypropyl methylcellulose (HPMC) and filled with compatible liquids or compositions that can flow below 70 oC.

Q: What are the advantages of liquid capsules?

A: Liquid-filled, two-piece capsules offer several advantages over other solid dosage forms, such as ease of scalability and manufacturing, faster absorption, simpler packaging, and higher product stability. Liquid filling also offers the ability to control R&D compounds, which are expensive and in limited supply.

Q: Are liquid capsules better than tablets?

A: WHAT WE FOUND Liquid and capsules contain the same amount of medicine and work equally well to treat an illness, according to medical researchers, but liquid medicine absorbs marginally faster than capsules.

Q: Do I chew liquid capsules?

A: The same rules apply for capsules and caplets. Look for a warning on the box before you attempt to chew them or cut them open. But this applies only to the solid forms of capsules. If you're taking a gelcap formulation (a soft, liquid-filled tablet), then it's never all right to chew or cut it open.

Q: Are liquid gels easier on your stomach?

A: Also known as liquid gels, they contain medication suspended in gelatin or a similar substance. This substance is easily digested, at which point active ingredients are released and absorbed.

Q: Why can't I swallow capsules?

A: There are a few reasons why you may have difficulty with medication in the form of pills, tablets, or capsules: A dislike for pills. Problems with the mechanics of swallowing. A temporary condition that makes it hard to swallow, such as taking antibiotics or going through chemotherapy.

Q: Do you drink water with capsules?

A: Water helps medicine pass from your mouth to your stomach and small intestine and to be absorbed to give the desired action. Swallowing medicines without enough water may prevent the medicine from acting properly and may even lead to undesired side effects in some cases.

Q: What are the benefits of liquid filled capsules?

A: Improved content uniformity of low-dose active and potent compounds. enhanced stability. lower moisture content. less gas transmission.

Q: Do capsules dissolve in your stomach?

A: Releasing the capsule's ingredients commencing when the capsule's shell degrades. Conventional gelatin hard capsules, enter the stomach in one to 5 min. The capsule's shell begins to degrade due to gelatin's sensitivity to the stomach's acidic environment and release the drug.

Q: What is the difference between capsules and hard capsules?

A: -Soft gelatin capsules normally hold liquids and semisolids while hard gelatin capsules frequently contain dry powders, plugs that have been slightly compressed, granules, or tablets. -While soft gelatin capsules come in round, oval, and tubular shapes, hard gelatin capsules are cylindrical.

Q: Do hard capsules dissolve in water?

A: Gelatin is used as the main ingredient of the hard capsules used in the pharmaceutical industry. An important property of these hard capsules is that they melt in water at a temperature above 30 °C and easily release drugs they contain in the human digestive tract due to temperature, gastric pH and digestive enzymes.

Q: How do hard capsules work?

A: These capsules are designed to resist heat and moisture, ensuring the stability of the contents during storage. Upon ingestion, the gelatin shell dissolves quickly in the acidic environment of the stomach, allowing for the rapid release of the active ingredients.

Q: Why are liquid capsules better?

A: Medical studies have shown that liquid extracts have faster absorption rates, higher optimisation rates, and are more easily digestible. Liquids may have a big advantage over pills when it comes to swallowing. Most people, especially the elderly and young children, tend to have more difficulty swallowing some pills.

Q: Is vitamin D better in liquid or capsule?

A: As far as potency and effectiveness, there is no significant difference among these dosage forms. However, the liquid vitamin may absorb quicker since it bypasses the intermediate step where the tablets have to be disintegrated by stomach acid before absorption.

Q: Can capsules get stuck in intestines?

A: A capsule that doesn't leave your body can cause a bowel obstruction, which can be serious. Healthcare providers do endoscopic surgery to remove capsules that don't leave the body naturally.
